The more people that play Gloomhaven the better. Because you have more characters it may make road and city events easier. Often having one of these characters makes for a favorable outcome so having 5 gives you an advantage.
Can I play Gloomhaven solo?
In the base game there are rules that allow you to play solo. You just are required to control two characters as that is the minimum per scenario in the base game. This is an additional set of solo quests to help boost your character. You can certainly play Gloomhaven solo without this.

How Long For a Full Campaign? Gloomhaven is a legacy board game and as such it has a story arc and persistent world changes. The campaign book has 95 scenarios, but to finish the game, while playing a decent amount of side quests, most groups end up playing approximately 65 scenarios.
